Description

Hexapeptide-11 is an anti-aging peptide that can protect fibroblasts from premature cell senescence mediated by oxidative stress. Hexapeptide-11 is also a potential stimulator of hyaluronic acid in keratinocytes, and can enhance skin barrier function by upregulating the expression of HAS2 and EGR3, and downregulating the expression of HYAL2. Hexapeptide-11 can also improve skin elasticity[1][2][3].

In Vitro

Hexapeptide-11 (2-5% v/v; 24-96 h) upregulates the expression of genes related to the proteostasis network in a dose- and time-dependent manner in human lung embryonic fibroblasts (IMR90) and human newborn foreskin fibroblasts (BJs), including genes related to proteasome, autophagy, chaperones, and antioxidant responses[1].
Hexapeptide-11 (5% v/v; 144 h) protects IMR90 cells against H2O2-mediated premature senescence and suppresses the upregulation of p53 and p21[1].
Hexapeptide-11 (1-5% v/v; 24 h) dose-dependently activates the enzymatic activity of MMP-2 and MMP-9 in IMR90 cells[1].
Hexapeptide-11 (1%-5% v/v; 24-72 h) can inhibit the migration activity of IMR90 cells[1].

Result: Increased the expression of 20S and 19S proteasome subunits genes (α7, β1, β2, β5, rpn6, rpn11).
Increased the expression of genes related to ALS functionality (i.e. becn1, sqstm1 and hdac6, along with the cathepsins ctsl and ctsd).
Resulted in the upregulation of molecular chaperones genes (hsf1, hsp27, hsp70, hsp90 and clu).
Increased the expression of genes related to cellular antioxidant responses, namely nrf2 and keap1, along with downstream transcriptional targets of Nrf2 (i.e. the nqo1 and txnrd1 genes).
