In Vitro

The antibody framework is stable, specific and adaptable, and has the ability to bind both antigens and endogenous immune receptors. Monoclonal antibodies have several derivatives, including bispecific antibodies, antibody-drug conjugates, and antibody fragments, and have significant effects in fields such as immunology and oncology. When designing inhibitory antibodies, considerations include identification of antigen-specific variable regions, choice of expression system, use of multispecific formats, and antibody derivatives based on fragmentation, oligomerization, or conjugation with other functional moieties[1].

In Vivo

Anti-Mouse IL-12 p40 Antibody (C17.8) (500 μg, i.p., twice a week during 5-30 weeks) exhibits suppression of both insulitis and diabetes in non-obese diabetic mice models [1].
Anti-Mouse IL-12 p40 Antibody (C17.8) (200 μg, i.p., for 6 consecutive days) results in the enhancement of diabetes in 2-week-old non-obese diabetic mice models[1].
Anti-Mouse IL-12 p40 Antibody (C17.8) (0.5 mg, i.p., 2 h before injection of SCW fragments) suppresses of murine streptococcal cell wall (SCW) arthritis[2].
Anti-Mouse IL-12 p40 Antibody (C17.8) (0.5 mg, i.p., once a week) prolongs survival in myelin oligodendrocyte glycoprotein-induced experimental autoimmune encephalomyelitis mice models[3].

Animal Model: Non-obese diabetic mice models[1]
Dosage: 500 μg
Administration: Intraperitoneally injection, twice a week during 5-30 weeks
Result: Completely prevented diabetes.
Significantly decreased the degree of insulitis.
Only 62% of NOD mice at 15 weeks of age.
Animal Model: 2-week-old non-obese diabetic mice models[1]
Dosage: 200 μg
Administration: Intraperitoneally injection, for 6 consecutive days
Result: Only 56% of NOD mice at 10 weeks of age.
Inhibited IL-2 and IFN-γ production at 4 weeks.
Enhanced production of IL-2 and IFN-γ and suppressed production of IL-4 and IL-10 at 8 weeks of age.
Increased CD25+ and CD44high cells in both CD4+ and CD8+ subsets at 8 days after the final injection.
Resulted in a rapid onset of overt diabetes when Splenic T-cells transferred to NOD-scid with spleen cells from diabetic NOD mice.
Animal Model: Streptococcal cell wall (SCW)-induced arthritis mice models[2]
Dosage: 0.5 mg
Administration: Intraperitoneally injection, 2 hours before injection of SCW fragments
Result: Increased the chondrocyte PG synthesis.
Reduced infiltration of synovial membrane with inflammatory cells.
Decreased mRNA levels for IL1β, IL12 and TNFα and increased IL1Ra and IL10 mRNA levels.
Animal Model: B6D2F1 mice were injected with 80 million B6 spleen cells[3]
Dosage: 0.5 mg
Administration: Intraperitoneally injection, every 7 days subsequently
Result: Significantly prolonged survival of mice.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
