Background

MICA Protein is devoid of a role in antigen presentation and functions as a stress-induced self-antigen recognized by gamma delta T-cells. Serving as a ligand for the KLRK1/NKG2D receptor, its binding to KLRK1 triggers cell lysis. Unlike classical MHC class I molecules, MICA does not form a heterodimer with beta-2-microglobulin but instead binds as a monomer to a KLRK1/NKG2D homodimer. KLRK1 forms a complex with HCST/DAP10, wherein KLRK1 binds MICA, and HCST acts as an adapter molecule facilitating signal transduction. Notably, MICA interacts with PDIA6 on the surface of tumor cells, leading to disulfide bond reduction necessary for the release of MICA from tumor cells. Additionally, MICA interacts with the human cytomegalovirus/HHV-5 protein UL142.
