GSDME Protein, Human (His) is the recombinant human-derived GSDME protein, expressed by E. coli, with N-6*His tag.

Background

DFNA5 is a precursor of a pore-forming protein that converts non-inflammatory apoptosis to pyroptosis. Its N-terminal moiety (Gasdermin-E, N-terminal) binds to membranes and forms pores upon cleavage, triggering pyroptosis. The protein can be cleaved by CASP3 or granzyme B (GZMB), promoting the switch from apoptosis to pyroptosis or granzyme-mediated pyroptosis, respectively. After cleavage, DFNA5 translocates to the plasma membrane, forms pores (10-15 nm in diameter), and releases mature interleukins (e.g., IL1B and IL16) to induce pyroptosis. It also binds to bisphosphorylated phosphatidylinositols, such as phosphatidylinositol (4,5)-bisphosphate. CASP3-mediated cleavage enables DFNA5 to convert TNF- or chemotherapy-induced apoptosis into pyroptosis. Additionally, DFNA5 mediates secondary necrosis downstream of mitochondrial apoptosis and viral infections, exhibits bactericidal activity, and suppresses tumors via GZMB-triggered pyroptosis in NK cell targets. Similar roles include: involvement in p53/TP53-regulated DNA damage response. Microbial infection-related function: DFNA5 promotes maternal placental pyroptosis during Zika virus infection, contributing to adverse fetal outcomes.
