Background

Deoxyhypusine synthase (DHS) plays a crucial role in the post-translational modification of the eIF-5A precursor protein. Catalyzing the NAD-dependent oxidative cleavage of spermidine, DHS subsequently transfers the butylamine moiety of spermidine to the epsilon-amino group of a lysine residue on the eIF-5A precursor. This process leads to the formation of the intermediate deoxyhypusine residue, representing the initial step in the conversion of the lysine into an atypical amino acid called hypusine. Hypusination is a unique modification specific to mature eIF-5A and is essential for its proper functioning.
