MCE Exosome Preservation Solution (Chemically Defined) is a buffer specifically formulated for the storage of exosome samples, designed to maintain their morphological integrity and biological activity. It is suitable for preserving exosomes at 4°C for up to 3 months or at -20°C for up to 6 months.

With a chemically defined composition, this product minimizes batch-to-batch variation and ensures accuracy and reproducibility in downstream analyses. It has been validated for key parameters including exosome integrity and particle size distribution, and is widely applicable for exosome isolation, transportation, storage, as well as subsequent functional or omics studies.

 

Feature of MCE Exosome Preservation Solution (Chemically Defined)

1. Endotoxin level strictly controlled to < 2.0 EU/mL, suitable for high-standard applications.

2. Chemically defined and mild formulation ensures stability of exosome structure and function.

3. Free of DMSO, glycerol, or other potentially interfering components.

4. Non-toxic to cells or animals when exosomes are used in downstream in vitro or in vivo studies.

Protocol

Sample Requirements

1. The exosome sample to be preserved should contain at least 3.00E+10 particles/mL to ensure proper visualization by TEM after addition of the preservation buffer.

2. For serum- or plasma-derived exosomes, NTA results are not recommended due to the high protein background. Instead, a TEM image of the original sample under 500 nm magnification showing at least three visible exosome particles is recommended before applying the preservation buffer.

 

Instructions for Use

1. Sample Preparation: Prepare exosome samples that meet the usage criteria.

2. Mixing with Preservation Buffer: Add an equal volume of the preservation buffer to the exosome sample (1:1, v/v), and mix gently by pipetting.

Note: Perform the operation in a biosafety cabinet. Ensure the preservation buffer remains sterile after opening. Seal the container with parafilm and store at 4°C to prevent degradation.

3. Aliquoting: Estimate the amount needed for single use and aliquot accordingly to facilitate future handling.

Note: Samples intended for -20°C storage should be aliquoted in advance to avoid repeated freeze-thaw cycles.

4. Storage: Store samples under appropriate conditions. At 4°C, samples are stable for up to 3 months; at -20°C, up to 6 months.

Note: For samples stored at -20°C, minimize freeze-thaw cycles and use or submit for analysis immediately after thawing.

Attention

1. The exosome sample must be prepared in strict accordance with the specified requirements to avoid potential interference from dilution by the preservation buffer in downstream analyses.

2. If the preserved exosomes are intended for subsequent cell-based or in vivo experiments, endotoxin levels in the original sample must be strictly controlled to ensure biosafety.

3. The preservation buffer should be opened in a biosafety cabinet. Keep the bottle neck clean and ensure the container is properly sealed during transfer to avoid microbial contamination and potential endotoxin elevation.

4. This product is for R&D use only, not for drug, household, or other uses.

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
