MCE Exosome-Specific Culture Medium (Serum-Free, Phenol Red-Free) is specifically formulated for optimized exosome collection. It is serum-free, phenol red-free, and completely xeno-free, allowing direct use without serum supplementation while supporting complete medium conditions. It is widely applicable for exosome production and research using tumor cells, T cells, and other cell types.

The formulation contains HEPES, L-Glutamine, pharm-grade albumin, hypoxanthine, thymidine, and glucose, providing stable support for cell growth and exosome secretion.

Free of growth factors, the medium minimizes exogenous interference and is favorable for downstream exosome purification and functional analysis. The absence of phenol red avoids estrogen-like effects on hormone-sensitive cells (e.g., breast cells) and reduces potential interference in flow cytometry and ion channel studies, improving the reliability and accuracy of experimental results.

Protocol

1. Cell Recovery and Expansion: After thawing, culture the cells in complete medium for 2-3 passages until they reach a stable growth state.

2. Phase I Adaptation: When cells reach approximately 50% confluency, remove the complete medium and replace it with a 1:1 mixture of Exosome-Specific Culture Medium (Serum-Free, Phenol Red-Free) and complete medium.

3. Phase II Adaptation: When cells grow to 70%-80% confluency in the phase I medium, discard the mixed medium.

4. Rinse the cells 2-3 times with PBS (1×) to remove residual components.

5. Add Exosome-Specific Culture Medium (Serum-Free, Phenol Red-Free) and continue culturing for 24-48 h. When cells reach 90%-100% confluency, collect the culture supernatant for downstream exosome isolation.

  • Do most proteins show cross-species activity?

    Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
