DDD2 is a selective and potent VHL-mediated PROTAC NUDT5 degrader. DDD2 induces robust NUDT5 degradation. DDD2 consists of the warhead TH10184 connected to the VHL recruiting ligand, VH032, through a saturated aliphatic eight carbon linker. In Vitro

DDD2 (2 µM, 24 h) induces robust NUDT5 degradation in U-2 OS cells[1].

DDD2 (0.01-10 µM, 4-96 h) induces concentration- and time-dependent degradation of endogenous NUDT5 in CCRF-CEM cells[1].

DDD2 (1 µM, 24 h) degrades NUDT5 depending on VHL (HY-P76124) not CRBN (HY-P702820) in U-2 OS cells[1].

DDD2 has good solubility and stability with Kinetic Solubility Mean of 6.25 µM[1].

Result: Induced robust NUDT5 degradation.

Concentration: 0.01, 0.03, 0.1, 0.3, 1, 3, and 10 µM
Incubation Time: 4, 24, and 96 h
