Description

BODIPY-cholesterol (solution) is an intrinsically lipophilic, and cell-permeable analog of cholesterol with a fluorescent BODIPY group. BODIPY-cholesterol can be used to monitor sterol uptake and inter-organelle sterol flux in cells. (Excitation/Emission: 505/515 nm)[1].

In Vitro

1. Solution preparation[1]
1.1 Stock solution
Storage: After aliquoting, store at -20°C or -80°C in the dark and avoid repeated freezing and thawing.
1.2 Preparation of working solution
Dilute with PBS (or other buffer). The corresponding stock solution can be diluted according to the actual situation. Note that if the solvent is DMSO, the cytotoxicity of DMSO must be considered, and a solvent control should be prepared; if the solvent is pure water, the working solution needs to be filtered and sterilized before adding cells.
Note: The working solution should be prepared and used immediately, and protected from light.

2. Cell staining
2.1 Wash cells with PBS.
2.2 Label the cell suspension with BODIPY-Cholesterol (final concentration 0.5-5 μM), mix well and incubate at 37°C for 10 -20 minutes.
2.3 Flow cytometric detection (Ex/Em = 510/590 nm).
