Background

SAM68 is an RNA-binding protein recruited and tyrosine-phosphorylated by multiple receptor systems (e.g., T-cell, leptin, and insulin receptors). Upon phosphorylation, it functions as an adapter in signal transduction by binding SH2/SH3 domain-containing proteins. It regulates G2-M cell cycle progression, represses CBP-dependent transcription by competing for CBP binding, and acts as a putative modulator of mRNA stability/translation and nuclear export. It enhances the association of CTE-containing mRNA with polyribosomes for translation initiation, though some studies exclude its role in exporting unspliced CTE-RNA. SAM68 binds RNA with a bipartite 5'-[AU]UAA-3' motif (spaced >15 nucleotides) and poly(A), influencing alternative splicing—e.g., CD44 (Ras-dependent), BCL2L1 (promoting Bcl-X(S) with HNRNPA1), and NRXN1/NRXN3 (via AS4 segment). Synergistic with KHDRBS2/SLIM-1 in activity-dependent NRXN1 exon skipping but antagonistic for NRXN3 (By similarity). Isoform 3, exclusive to growth-arrested cells, inhibits S-phase progression.

  • Western blot analysis of extracts from Hela (lane 2(20μg), Hela (lane 3(40μg), using SAM68 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded mouse kidney tissue using SAM68 Antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
