FGFR1 Antibody (YA441) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to FGFR1.

FGF Receptor 1 is a tyrosine-protein kinase that functions as a cell-surface receptor for fibroblast growth factors (FGFs) and plays essential roles in regulating embryonic development, cell proliferation, differentiation, and migration. It is required for proper mesoderm patterning and axial organization during embryogenesis, normal skeletogenesis, and development of the gonadotropin-releasing hormone (GnRH) neuronal system. This receptor phosphorylates substrates including PLCG1, FRS2, GAB1, and SHB. Upon ligand binding, FGFR1 activates multiple signaling cascades: PLCG1 activation generates secondary messengers diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3); FRS2 phosphorylation mediates recruitment of GRB2, GAB1, PIK3R1, and SOS1 to activate RAS-MAPK (MAPK1/ERK2, MAPK3/ERK1) and AKT1 pathways; it also promotes phosphorylation of SHC1, STAT1, and PTPN11/SHP2. In the nucleus, FGFR1 enhances RPS6KA1 and CREB1 activity to regulate transcription. Its signaling is negatively regulated by IL17RD/SEF and through ubiquitination, internalization, and degradation mechanisms.

  Western blot analysis of extracts from SH-SY5Y(lane 2(20μg) or lane 3(20μg)) using FGFR1 alpha (HY-P80395) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80395, 1/1000) and Loading control antibody (GAPDH, HY-P80954, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
