Doublecortin Antibody (YA3541)

Doublecortin Antibody (YA3541) is a mouse-derived and non-conjugated IgG1 antibody, targeting to DCX. It can be applicated for WB, IHC-P, ICC/IF, FC, ELISA assays, in the background of human, mouse, rat, rabbit, monkey.

DCX is a microtubule-associated protein essential for the initial steps of neuronal dispersion and cortex lamination during cerebral cortex development. It may function by competing with the putative neuronal protein kinase DCLK1 in binding to a target protein, thereby participating in a signaling pathway critical for neuronal interaction before and during migration, possibly as part of a calcium ion-dependent signal transduction pathway. Additionally, DCX may collaborate with PAFAH1B1/LIS-1 in overlapping yet distinct signaling pathways that promote neuronal migration.

  Western blot analysis of extracts from SH-SY5Y (lane 2(20μg), SH-SY5Y (lane 3(40μg), using DCX Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

  Immunohistochemical analysis of paraffin-embedded mouse kidney tissue using DCX Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
