Description

CELF2 Antibody (YA2480) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to CELF2.

Background

CELF2 is an RNA-binding protein involved in regulating various post-transcriptional events, including pre-mRNA alternative splicing, mRNA translation, and stability. It mediates tissue-specific and developmentally regulated splicing, such as specifically activating TNNT2 exon 5 inclusion in embryonic skeletal muscle by antagonizing PTB's repressive effect. CELF2 functions as both an activator and repressor for coregulated exon pairs, promoting smooth muscle (SM) exon inclusion while excluding non-muscle (NM) exon in actinin pre-mRNAs. It also participates in NMDA receptor R1 pre-mRNA exon 21 inclusion and exon 5 exclusion, as well as apoB RNA editing. Post-radiation injury, CELF2 enhances COX2 mRNA stability while inhibiting its translation (by similar) and modulates cellular apoptosis by regulating COX2-mediated prostaglandin E2 (PGE2) expression (by similar). CELF2 binds diverse RNA sequences, including (CUG)n repeats in DMPK 3'-UTR, muscle-specific splicing enhancer (MSE) intronic sites flanking TNNT2 exon 5, UG-rich motifs (e.g., UG repeats and UGUU), AU-rich sequences upstream of apoB's edited cytidine (by similar), and AU-rich elements in COX2 mRNA 3'-UTR (by similar). Additionally, it may regulate miRNA biogenesis, such as binding pri-MIR140 with CELF1 to suppress mature miRNA processing.

  • Western blot analysis of extracts from Jurkat(lane 2(20ug) , THP-1(lane 3(20ug) and HEK293(lane 4(20ug) using CELF2 Antibody (HY-P82735) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded Mouse colon tissue using CELF2 Antibody (YA2480). The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P82735, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
