Description

Xentry is a cell-penetrating peptide (CCP) consisting of only 7 amino acids of hepatitis B virus: LCLRPVG. Xentry-linked anti-B-raf antibodies and siRNAs demonstrates the capability to kill B-raf-dependent melanoma cells. Xentry alone or conjugated to β-galactosidase leads to its delivery to most tissues in mice, except circulating blood cells. Xentry can be used for the delivery of large molecules (antibodies, siRNA, enzymes) [1].

In Vitro

Xentry (10 μM) penetrates adherent cells energy-dependently, including HepG2 (liver), C32 (melanoma), DU145 (prostate), H441 (lung), BT474 (breast), Cos (kidney), and Rin-m5F (pancreatic b-cell) cell lines, but not nonadherent resting blood cells[1].
Xentry (10-20 μM, 4 h) conjugated to β-galactosidase is internalized by living HepG2 cells, and enters cells via a syndecan pathway[1].
Xentry (1 h) effectively delivers antibodies and siRNA to melanoma cells, inducing apoptosis of B-raf-dependent melanoma cells[1].

In Vivo

Cy7-labelled Xentry (2-20 μg/mL, i.v., single dose) is widely distributed in various tissues within 15 minutes, with a preferential accumulation in epithelial tissues (intestine, stomach, bronchus), and mainly remains in the liver and kidneys after 24 hours in Balb/c mice[1].
TAMRA-labelled Xentry (2-100 μg/mL, i.v., single dose) successfully delivers the macromolecular complex to multiple tissues in Balb/c mice, specifically enriching in small intestinal epithelial cells and marginal sinus of the spleen[1].
