Description

Sulfo Cy7 tyramide, a red fluorescent dye (Ex=740 nm, Em=770 nm), is utilized as reporter fluorescent substrate of horseradish peroxidase (HRP)-catalyzed deposition for tyramide signal amplification (TSA). Sulfo Cy7 tyramide can be used for multiplex Immunohistochemistry (mIHC)[1].

In Vitro

Sulfo Cy7 tyramide assay
a. Cells are fixed with 3.7% paraformaldehyde in 1x PBS at RT for 20 minutes, and then rinse with 1x PBS twice.
b. permeabilize in 0.1% Triton X-100 for 1-5 minutes at RT, and then rinse with 1x PBS twice.
c. Incubate in 3% H2O2 in 1x PBS for 10 min to block endogenous peroxidase activity, and then rinse with 1x PBS twice.
d. Incubate in 1x PBS plus 1% BSA for 30 min at 4℃.
e. Apply the primary antibodies working solution for 1 h at RT or overnight at 4 °C, and then rinse with 1x PBS three times, 5 minutes each.
f. Apply 100 µL secondary antibody-HRP working solution for 1 h at RT and then rinse with 1x PBS three times, 5 minutes each.
g. Incubate in 100 µL Sulfo Cy7 tyramide working solution for 5-10 minutes at RT, and then rinse with 1x PBS three times, 5 minutes each.
h. Fluorescence microscopy detection.
Note: it is suggested that the optimal concentration of Sulfo Cy7 tyramide working solution according to actual conditions.
