Description

SQS-IN-1 is a squalene synthase (SQS) inhibitor. SQS-IN-1 exhibits potent and broad-spectrum anti-proliferative effects on both mouse and human lung cancer cell lines. SQS-IN-1 inhibits DNA replication and the cell cycle, causing mitochondrial hyperpolarization and inducing cell apoptosis. SQS-IN-1 inhibits cell migration and invasion. SQS-IN-1 can be used to the study of lung cancer[1].

In Vitro

SQS-IN-1 (Compound 2) (0.1 nM-100 μM, 24-72 h) inhibits various lung cancer cell lines proliferation including LLCOVA cells, NSCLC cell lines (H1299 and H358) and B16 melanoma cells and with TC50s of 3.21 nM, 1.6 μM, 0.55 μM and 0.21 μM at 72 h[1].
SQS-IN-1 (10 and 0.1 μM, 24 h) induces G0/G1 phase arrest in A549 cells and arrested DNA replication in LLC-OVA cells[1].
SQS-IN-1 (0.1 μM, 24 h) induces LLC-OVA cells apoptosis and effectively inhibits the metastasis ability of cancer cells[1].
SQS-IN-1 (0-10 μM) causes mitochondrial hyperpolarization in a dose-dependent manner in LLC-OVA cells[1].

Cell Cycle Analysis[1]

Cell Line: A549 and LLC-OVA cells
Concentration: 10 μM (A549) and 0.1 μM (LLC-OVA)
Incubation Time: 24 h
Result: Significantly reduced the proportion of cells in the S phase and the G2 phase, resulting in the accumulation of cells in the G0/G1 phase in A549 cells.
Inhibited DNA replication and the cell cycle progression in LLC-OVA.
