Description

RX 336M is a dihydrocodeinone analogue. RX 336M induces behaviors consisting of consisting of behavioral activation, rapid forepaw movements, excessive grooming, and whole body "wet-dog" shakes (WDS) (rapid twisting of the head and trunk with the forepaws leaving the ground). RX 336M can be used for the study of neurological disease[1][2].

In Vivo

RX 336M (1.5-12 mg/kg, i.p., single dose) induces dose-, age- and sex-related "wet-dog" shakes and excessive grooming in rats[1][2].

Animal Model: Male Sprague Dawley rats (weight ranges: 80-90 g, 120-140 g, 190-210 g, 280-300 g)[1]
Dosage: 1.5, 3, 6, 12 mg/kg
Administration: i.p. for a single dose
Result: Induced grooming behavior that primarily involved face washing and preening of the flanks with the mouth or forepaws, with very little hindpaw scratching.
Caused a sequential behavioral pattern in rats: shaking first, followed by face washing, then grooming of the flanks, and repetition of this sequence.
Led to a decrease in the correlation between the number of "wet-dog" shakes (WDS) and the percentage of maximum possible grooming episodes (%MGE).
Showed improved correlation between WDS and %MGE in rats weighing 190-210 g and 280-300 g.
Increased the number of WDS but decreased the correlation between WDS and grooming when the dose was doubled to 12 mg/kg in large rats.
Resulted in a decline in %MGE but an improvement in the correlation between shaking and grooming.
Animal Model: Male and female Long-Evans rats of different ages (21 days, 30 days, 39 days, and 48 days old)[2]
Dosage: 6 mg/kg
Administration: i.p. for a single dose
Result: Induced more significant excessive grooming in younger rats than in older ones, with gender differences.
Caused age- and sex-related differences in the frequency of "wet-dog" shakes (WDS).
Had no significant effect on the activity levels (rearing and locomotion) of rats, while female rats generally had higher activity levels than males.
Animal Model: 39-day-old female Long-Evans rats received daily subcutaneous injections of testosterone benzoate (1 mg/kg/day) for 8 consecutive days starting from 32 days old[2]
Dosage: 6 mg/kg
Administration: i.p. for a single dose
Result: Enhanced the responses of female rats to excessive grooming and "wet-dog" shakes.
Had no significant behavioral effects in 39-day-old female rats pretreated with oil vehicle.
