Background

Slit2, a pivotal protein in cellular migration, acts as a molecular guidance cue with its function mediated by interactions with roundabout homolog receptors. In neural development, Slit2 contributes to axonal navigation at the ventral midline of the neural tube, guiding the projection of axons to distinct regions. Particularly essential in forebrain midline guidance, SLIT1 and SLIT2 act as repulsive signals, preventing improper midline crossing by axons originating from the olfactory bulb. In spinal cord development, Slit2 likely plays a role in guiding commissural axons upon reaching the floor plate, modulating responses to netrin. Silencing the attractive effect of NTN1 in vitro requires the formation of a ROBO1-DCC complex. Furthermore, Slit2 is implicated in spinal cord midline post-crossing axon repulsion. In the developing visual system, it serves as a repellent for retinal ganglion axons, directing them along appropriate paths before and after the optic chiasm. Slit2 is also involved in branching and arborization of CNS sensory axons and neuronal cell migration. Notably, Slit2 regulates leukocyte migration, adding to its diverse functions. Interacting with GREM1, Slit2 forms homodimers and binds to ROBO1 and ROBO2 with high affinity.
