PYK2 is a non-receptor protein-tyrosine kinase that orchestrates actin cytoskeleton reorganization, cell polarization, migration, adhesion, spreading, and bone remodeling. It modulates humoral immunity by maintaining marginal zone B-cell populations in the spleen and enabling B-cell trafficking, while being essential for macrophage polarization and chemotaxis toward inflammatory sites. In T-cells, it regulates cytoskeletal rearrangement and contributes to immune responses. Collaborating with SRC, it enhances osteoclastic bone resorption and may suppress osteoprogenitor differentiation. Functioning downstream of diverse receptors (integrins, GPCRs, cytokine/chemokine receptors) and stress signals, it forms activated signaling complexes with SRC family kinases to create phosphotyrosine docking sites. Its multifaceted signaling includes PI3K-AKT1 activation, NOS3 stimulation, cGMP production, MAPK cascade initiation (ERK1/2, JNK1), and Rho GTPase (RHOA/RAC1) regulation. Nuclear PYK2 recruits MDM2 to modulate p53 ubiquitination/degradation, and acts as a scaffold for SRC-mediated PDPK1 phosphorylation. It influences NMDA receptor activity via SRC-family phosphorylation, potentially regulates potassium channels, and auto-amplifies SRC kinase activity. Substrates include ASAP1, NPHP1, KCNA2, SHC1, with SRC-cooperative phosphorylation of ASAP2, RHOU, and PXN.
