Periodic Acid-Silver Methenamine Staining Kit for Basement Membrane can be used for staining basement membrane mucopolysaccharides. Its staining principle is that after tissue is oxidized by iodic acid, the mucopolysaccharides within the basement membrane expose aldehyde groups. These aldehyde groups reduce hexamine silver to black metallic silver, and gold chloride transforms the metallic silver into a more stable form of gold.

Periodic Acid-Silver Methenamine Staining Kit for Basement Membrane can be used for staining basement membrane mucopolysaccharides. Its staining principle is that after tissue is oxidized by iodic acid, the mucopolysaccharides within the basement membrane expose aldehyde groups. These aldehyde groups reduce hexamine silver to black metallic silver, and gold chloride transforms the metallic silver into a more stable form of gold.

In Vitro

Guidelines (The following are our recommended protocols. This protocol is for reference only and should be modified according to your specific needs.)
1. Dewax sections with xylene or wax-removing and clearing solution until dissolved in water.
2. Oxidize sections in periodic acid solution for 15 minutes, then wash with tap water and distilled water for 2 minutes each.
3. Stain sections in iron alum solution for 10 minutes, then wash with tap water and distilled water for 2 minutes each.
4. Prepare hexamine silver stock solution: silver solution: hexamine solution = 6:100. A small amount of precipitate may form; dissolve thoroughly.
5. Prepare hexamine silver working solution: hexamine silver stock solution: borax solution = 106:100. The hexamine silver working solution is for single use only and should not be stored for long periods. After preparation, preheat in a 60°C incubator for 30 minutes before staining the sections.
6. Immerse the sections in preheated hexamine silver working solution and stain in a 60°C water bath for approximately 25-30 minutes, until the sections turn tobacco yellow or black. Rinse with distilled water.
7. Adjust the color with gold chloride solution for 1-2 minutes, wash with distilled water for 2 minutes, treat with sodium thiosulfate solution for 1 minute, and rinse with tap water.
8. Counterstain with a pale green solution for 1 minute, rinse with tap water, dehydrate rapidly with 95% ethanol, clear with xylene or wax-removing and clearing solution, and mount with neutral resin.

Staining Results:
The glomerular capsule basement membrane and renal capillary glomerular basement membrane appear black;
The background appears green.

Notes:
1. Glassware used in the experiment should be pre-soaked in washing solution (if washing solution is unavailable, it can be soaked in 0.1 M hydrochloric acid and rinsed thoroughly), rinsed clean, and dried.
2. Hexamine silver stock solution can be stored at 4°C and is effective for 3 months.
3. Hexamine silver working solution is for single use only and should not be stored for extended periods.
4. When adjusting the color of gold chloride, it is essential to observe it under a microscope and monitor the color changes in real time.
