Oil Red O Staining Kit (For Slides) 

Oil Red O is a fat-soluble diazol dye, with a maximum absorption at 518 nm. Oil Red O stains neutral lipids and cholesteryl esters but not biological membranes. Oil Red O Staining Kit (For Cultured Cell) can be used for staining cell smears, bone marrow smears, body fluid smears, blood smears, etc.

Guidelines (The following are our recommended protocols. This protocol is for reference only and should be modified according to your specific needs.)
1. Prepare fresh bone marrow, blood, and cell smears and fix them on Cell ORO Fixative for 10-15 min.
2. Remove the smears and place them in circulating air for 10-15 min.
3. Stain with Oil Red O staining solution for 10-15 min.
4. Wash with 60% isopropanol for 20-30 s, then rinse with running water.
5. Counterstain cell nuclei with hematoxylin staining solution for 2-5 min.
6. Wash with ORO Buffer for 1 min (optional).
7. Wash with distilled water for 8-10 min.
8. Air dry and examine under a microscope.

Staining results:
Neutral fats appear orange-red or tangerine-red;
Phospholipids appear pink;
Cell nuclei appear blue.

Note:
1. ORO staining solution should be prepared fresh before use.
2. Staining results cannot be stored for long periods and should be observed and photographed as soon as possible.
