Description

ODN 1826 is a class B CpG ODN (oligodeoxynucleotide) and TLR9 agonist. ODN 1826 induces NO and iNOS production and enhances Apoptosis. ODN 1826 enhances immune surveillance. ODN 1826 increases aortic atherosclerotic plaque size. ODN 1826 has antitumor activity against lung cancer, glioma and melanoma[1][2][3][4][5][6].

In Vitro

ODN 1826 (1 μg/mL; 24 h) induces nitric oxide (NO) and inducible nitric oxide synthase (iNOS) production in RAW 264.7 mouse macrophages[3].

In Vivo

CpG-1826 (100 μg/tumor; intratumoral injection; every 3 days for up to 4 times) delays the growth of subcutaneously implanted GL261 gliomas in C57BL/6NTac mice, inducing increased infiltration of macrophages and B cells into tumors, with significant growth delay in ~50% of mice[4]. ODN 1826 (18 nM; s.c.; 3 times a week for 7 weeks) increases aortic atherosclerotic plaque size in a chronic vascular injury mouse model[5].
ODN 1826 (0.05 mg; i.p.; 1, 3, 5, 8, 11, 13 days) has a strong anti-tumor growth effect in a Lewis lung cancer mouse tumor model[6].

Animal Model: 6-week-old male C57BL/6NTac mice; GL261 glioma or B16F10 melanoma subcutaneously implanted in posterior flanks[4].
Dosage: 100 μg/tumor
Administration: Intratumoral injection; every 3 days for up to 4 treatments
Result: Slowed tumor growth, increased F4/80+ macrophages and B220+ B cells in tumors, with variable responses (50% mice showed prolonged stasis) (in GL261 glioma model).
Achieved complete tumor regression and induced long-term immune memory in combination with Cyclophosphamide (HY-17420).
Delayed growth but did not induce regression, with modest increases in F4/80+ macrophages and B220+ B cells in combination with Cyclophosphamide (in B16F10 melanoma model).
