Description

Hydrostatin-TL1 (H-TL1), a nine-amino-acid peptide, is a TNF-α antagonist. Hydrostatin-TL1 can be isolated from the venom gland of sea snake Hydrophis cyanocinctus. H-TL1 competitively inhibits the interaction between TNF-α/TNFR1 and attenuates the cytotoxicity and inflammation of TNF-α. In Vitro

Hydrostatin-TL1 (1-1000 nM, 18-20 h) inhibits TNF-α-induced L929 cells death in a concentration-dependent manner [1].
Hydrostatin-TL1 (0.2-1.6 μM) significantly inhibits the TNF-α-induced phosphorylation of p38, ERK, JNK, and IκB protein within the NF-κB and MAPKs signaling pathways in HEK-293 cells[1].
Hydrostatin-TL1 fails to inhibit the phosphorylation of IκB induced by IL-1β in HEK-293 cells[1].
Hydrostatin-TL1 (0.1-0.8 nM) dose-dependently reduces TNF-α-mediated mRNA expression of pro-inflammatory factors (IL-8, TNF-α, ICAM-1, and VCAM-1) in HEK-293 cells[1].

In Vivo

Hydrostatin-TL1 (0.5-1 mg/kg, i.p., daily for 7-8 days) has significant anti-inflammatory activity against DSS (HY-116282C)-induced acute colitis mice model[1].
Hydrostatin-TL1 (20-80 μg/kg, three times at 1, 3 and 5 h) dose-dependently reduces mortality in LPS (HY-D1056)-induced acute shock mice model[1].
