DNA polymerase-IN-7 is a noncompetitive inhibition of DNA Polymerase β with an IC50 of 112 nM and a Ki of 35 nM. DNA polymerase-IN-7 can inhibit DNA pol β lyase activity and the binding of the lyase domain of Pol β to DNA. DNA polymerase-IN-7 can be used for the research of cancer, such as cervical cancer[1].

In Vitro

DNA polymerase-IN-7 (Compound 28) (0.1-1 μM) inhibits dGTP incorporation activity of DNA polymerase β (Pol β) and inhibits the lyase activity of Pol β[1].
DNA polymerase-IN-7 (0.35-2 μM) exerts little effect on the binding of Pol β to DNA, but inhibits the binding of the lyase domain of Pol β to DNA[1].
DNA polymerase-IN-7 (0.1-1 μM) exhibits selectivity for X-family polymerases, with inhibitory rate of 50% for Pol β and 41% for Pol λ at 0.1 μM, and almost 100% at 1 μM[1].
DNA polymerase-IN-7 (0-100 μM, 24 h) shows no obvious cytotoxicity, maintaining cell viability at 95-100% in HeLa cells[1].
DNA polymerase-IN-7 (25 μM, 24 h) shows intracellular concentration reaching 4.0 μM in HeLa cells[1].
