Description

RANKL Antibody (YA3508) is a mouse-derived and non-conjugated IgG1 antibody, targeting to TNFSF11. It can be applicated for WB, IHC-P, ICC/IF, FC, ELISA assays, in the background of human, mouse, rat, monkey, rabbit.

Background

TNFSF11 is a cytokine that binds to TNFRSF11B/OPG and TNFRSF11A/RANK, functioning as an osteoclast differentiation and activation factor. It enhances the ability of dendritic cells to stimulate naive T-cell proliferation and may serve as a key regulator of T-cell and dendritic cell interactions, playing a role in T-cell-dependent immune response regulation. Additionally, TNFSF11 may contribute to increased bone resorption in humoral hypercalcemia of malignancy. It induces osteoclastogenesis by activating multiple signaling pathways in osteoclast precursor cells, notably triggering prolonged oscillations in intracellular Ca 2+ concentration, leading to NFATC1 activation and nuclear translocation to drive osteoclast-specific gene transcription. During osteoclast differentiation, it promotes CREB1 activation and mitochondrial ROS generation in a TMEM64- and ATP2A2-dependent manner, essential for proper osteoclast formation (By similarity).
