Background

SV40 T Antigen is a pivotal early protein essential for driving viral replication and inducing cellular transformation. It facilitates viral genome replication by triggering quiescent cells to enter the cell cycle and autoregulating viral early mRNA synthesis. Its highly oncogenic activities stem from subverting host cellular checkpoint mechanisms governing cell division, DNA transcription, replication, and repair. Prior to viral DNA replication, it modulates cellular gene expression by binding key host cell cycle regulators RB1/pRb and TP53. It induces E2F1 transcription factor dissociation from RB1, activating E2F1-regulated S-phase genes, while inhibiting TP53's DNA-binding and transcriptional activation functions. Acting as a TFIID-associated factor (TAF), it stabilizes the TBP-TFIIA complex on promoters to initiate transcription by all three RNA polymerases. It initiates viral DNA replication and unwinding by interacting with the SV40 origin, with its helicase activity promoting replication fork progression. The protein exhibits processive 3'-5' DNA helicase activity requiring ATP and a 3' single-stranded region (other (d)NTPs partially substitute ATP). It activates viral late mRNA transcription via host TBP/TFIIA stabilization and disrupts HDAC1-mediated histone deacetylation to stimulate transcription. It may inactivate the growth-suppressing E3 ubiquitin ligase CUL7. The 17kT isoform degrades host RBL2 to promote proliferation and transactivates the cyclin A promoter via its J domain. Additionally, it unwinds G4 DNA (hydrogen-bonded planar guanine quartets, tested in parallel/antiparallel configurations) in a 3'-5' direction, dependent on a 3' single-stranded tail and hydrolyzable ATP.
