SP7/Osterix Antibody (YA3539)

SP7/Osterix Antibody is an unconjugated, approximately 45 kDa, rabbit-derived, anti-SP7/Osterix monoclonal antibody. SP7/Osterix Antibody can be used for: WB, IHC-P, IF-Tissue, IHC-Fr expriments in human, mouse, and rat background without labeling.

The related gene encodes a member of the Sp subfamily of Sp/XKLF transcription factors. Sp family proteins are sequence-specific DNA-binding proteins characterized by an amino-terminal trans-activation domain and three carboxy-terminal zinc finger motifs. The protein is a bone specific transcription factor and is required for osteoblast differentiation and bone formation.

  Western blot analysis of extracts from Saos-2 (lane 2(20μg) , Saos-2 (lane 3(40μg) ,using SP7/Osterix Antibody (HY-P81125). Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

  Immunohistochemical analysis of paraffin-embedded Rat brain tissue using SP7/Osterix Antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P81125, 1/500) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
