Smad4 Antibody (YA6044) is a rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Smad4. It can be applicated for WB, IHC-P, ICC/IF, IP, ELISA assays, in the background of human, mouse, rat.

Smad4 plays a central role in muscle physiology by regulating the balance between atrophy and hypertrophy. When recruited by MSTN, it promotes atrophy via phosphorylated SMAD2/4, while decreased MSTN leads to Smad4 release and subsequent recruitment by the BMP pathway to induce hypertrophy through phosphorylated SMAD1/5/8. It acts synergistically with SMAD1 and YY1 in BMP-mediated cardiac-specific gene expression and binds to SMAD binding elements (SBEs, 5'-GTCT/AGAC-3') within the BMP response element (BMPRE) of cardiac activating regions (By similarity). As a common SMAD (co-SMAD), Smad4 serves as a coactivator and mediator of TGF-beta signaling. It is a component of the heterotrimeric SMAD2/SMAD3-SMAD4 complex required for TGF-mediated signaling. Smad4 facilitates DNA binding of the SMAD2/SMAD4/FAST-1 complex and provides transcriptional activation function for SMAD1 or SMAD2. It also participates in the multimeric SMAD3/SMAD4/JUN/FOS complex at the AP1 promoter site, enabling synergistic transcriptional activity in response to TGF-beta. Smad4 may act as a tumor suppressor and positively regulates PDPK1 kinase activity by promoting its dissociation from the inhibitory 14-3-3 protein YWHAQ.
