Raf-1 is a serine/threonine-protein kinase that serves as a critical regulatory link between membrane-associated Ras GTPases and the MAPK/ERK cascade, functioning as a molecular switch governing cell fate decisions such as proliferation, differentiation, apoptosis, survival, and oncogenic transformation. Its activation initiates a MAPK cascade involving sequential phosphorylation of dual-specific MAPK kinases (MAP2K1/MEK1 and MAP2K2/MEK2) and extracellular signal-regulated kinases (MAPK3/ERK1 and MAPK1/ERK2). Raf-1 phosphorylates diverse substrates including BAD (at Ser-75), adenylyl cyclases (ADCY2/5/6), PPP1R12A (inhibiting its phosphatase activity), and cardiac troponin T (TNNT2). It regulates cell survival through multiple mechanisms: 1) translocating to mitochondria to bind BCL2 and displace BAD; 2) modulating NF-κB activation and inhibiting signaling molecules like ROCK2, MAP3K5/ASK1, STK3/MST2, and RB1 to influence motility, apoptosis, proliferation, and angiogenesis; 3) promoting epithelial cell transformation by upregulating transcriptional repressor SNAI2/SLUG to downregulate tight junction protein occludin (OCLN); and 4) restricting caspase activation in response to specific stimuli including Fas stimulation, pathogen-induced macrophage apoptosis, and erythroid differentiation.
