R-Spondin Antibody (YA5049) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to R-Spondin.

R-Spondin is a protein that activates the canonical Wnt signaling pathway by acting as a ligand for LGR4-6 receptors (LGR4, LGR5, or LGR6). Upon binding to LGR4-6, these receptors associate with phosphorylated LRP6 and frizzled receptors, which are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase target gene expression. Additionally, R-Spondin regulates the canonical Wnt/β-catenin-dependent pathway and non-canonical Wnt signaling by inhibiting ZNRF3, a key regulator of the Wnt signaling pathway. It also acts as a ligand for frizzled FZD8 and LRP6 and may negatively regulate the TGF-β pathway. R-Spondin plays an essential role in ovary determination. By interacting with KREM1, it antagonizes DKK1/KREM1-mediated internalization of LRP6, thereby modulating Wnt signaling.
