NCOA4 is a cargo receptor for the autophagic turnover of the iron-binding ferritin complex, playing a central role in iron homeostasis. It acts as an adapter to deliver ferritin to lysosomes for autophagic degradation, a process termed ferritinophagy. Under conditions of starvation or iron depletion, NCOA4 targets the ferritin complex to autolysosomes. It ensures efficient erythropoiesis, potentially by regulating hemin-induced erythroid differentiation. Some studies indicate that NCOA4 enhances androgen receptor (AR) transcriptional activity and functions as a ligand-independent coactivator of peroxisome proliferator-activated receptor (PPAR) gamma; however, another study reports only weak coactivator behavior for AR without altering its ligand responsiveness. NCOA4 also binds to DNA replication origins, independent of active transcription sites and likely unrelated to its nuclear receptor coactivator function, and may inhibit replication origin activation by obstructing DNA unwinding via interaction with the MCM2-7 complex.
