Background

MUC4 is a membrane-bound mucin, a family of highly glycosylated proteins that form the major component of mucus covering epithelial surfaces. These glycoproteins play vital roles in epithelial protection, renewal, and differentiation. MUC4 regulates cellular behavior through anti-adhesive effects on cell-cell and cell-extracellular matrix interactions while acting as an intramembrane ligand for ERBB2. It critically influences epithelial cell proliferation and differentiation by inducing ERBB2-specific phosphorylation. In polarized epithelial cells, MUC4 segregates ERBB2 from other ERBB receptors, preventing its coreceptor function. Interaction with ERBB2 enhances CDKN1B expression, whereas the MUC4-ERBB2-ERBB3-NRG1 complex downregulates CDKN1B, suppressing apoptosis and stimulating proliferation. Its pro-tumorigenic effect likely stems primarily from apoptosis repression rather than direct proliferation promotion.
