mTOR Antibody (YA281) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to mTOR.

Background

mTOR is a serine/threonine protein kinase that serves as a central regulator of cellular metabolism, growth, and survival in response to hormones, growth factors, nutrients, energy, and stress signals. It functions through two structurally and functionally distinct complexes, mTORC1 and mTORC2. mTORC1 is recruited to the lysosome membrane upon nutrient or growth factor stimulation, promoting protein, lipid, and nucleotide synthesis by phosphorylating key regulators of translation (e.g., EIF4EBP1, RPS6KB1/2) while suppressing autophagy (e.g., via ULK1, DAP, AMBRA1 phosphorylation). mTORC2 responds to growth factors by phosphorylating AGC kinase family members (e.g., AKT, PKC, SGK1) to regulate proliferation, cytoskeletal organization, and lipogenesis. Additionally, mTOR modulates mitochondrial biogenesis (by similar), insulin signaling (by similar), circadian rhythms (by similar), and pyroptosis (e.g., GSDMD oligomerization).

  • Western blot analysis of extracts from Hela (lane 2(20μg), HepG2 (lane 3(20μg), HEK-293 (lane 4(20μg) using mTOR Antibody (HY-P80231) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (HY-P80231, 1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded Rat kidney tissue using mTOR Antibody (YA281). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80231, 1/500) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
