MICU1 Antibody (YA5426) is a mouse-derived and non-conjugated monoclonal antibody, targeting to MICU1. It can be applicated for WB, ICC/IF, IHC-P assays, in the background of human, mouse, rat.

MICU1 is a calcium sensor of the mitochondrial calcium uniporter (MCU) channel, sensing calcium levels via its EF-hand domains. It forms a disulfide-linked heterodimer with MICU2 (or MICU3) that stimulates or inhibits MCU activity depending on calcium concentration. At low calcium levels, MICU1 occludes the MCU pore to prevent mitochondrial calcium uptake, while at higher calcium levels, calcium binding induces a conformational change that weakens MCU-MICU1 interactions and moves the MICU1-MICU2 heterodimer away from the pore, enabling calcium permeation. Additionally, MICU1 protects against manganese toxicity by blocking MCU-mediated manganese uptake, as manganese binding to its EF-hands does not trigger conformational changes, maintaining pore occlusion. It also acts as a barrier for MCU inhibitors like ruthenium red or Ru360 and regulates mitochondrial cristae structure independently of MCU modulation. In pancreatic beta-cells, MICU1 controls glucose-dependent insulin secretion by regulating mitochondrial calcium uptake and induces Th1-mediated autoreactivity accompanied by IFNG release. By similarity: This isoform regulates MCU in skeletal muscle and exhibits higher calcium affinity than other isoforms, promoting mitochondrial calcium uptake at lower concentrations to rapidly adapt metabolism and sustain ATP production during resistance or strenuous exercise.
