JAB1 Antibody (YA3159) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to JAB1.

JAB1 is a probable protease subunit of the COP9 signalosome complex (CSN), which is involved in various cellular and developmental processes. The CSN complex acts as an essential regulator of the ubiquitin (Ubl) conjugation pathway by mediating the deneddylation of cullin subunits in SCF-type E3 ligase complexes (e.g., SCF, CSA, or DDB2), thereby reducing their Ubl ligase activity. The complex may also participate in the phosphorylation of p53/TP53, c-jun/JUN, IkappaBalpha/NFKBIA, ITPK1, and IRF8, potentially through its association with CK2 and PKD kinases. CSN-dependent phosphorylation of TP53 and JUN promotes and protects their degradation by the Ubl system, respectively. Within the complex, JAB1 likely serves as the catalytic center for cleaving Nedd8 from cullins, though it lacks intrinsic metalloprotease activity and relies on other CSN subunits. JAB1 directly interacts with numerous proteins regulated by the CSN complex, highlighting its pivotal role, and promotes the proteasomal degradation of BRSK2.

  Immunocytochemistry analysis of A549 cells labeling JAB1 with JAB1 Antibody (HY-P83414) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with JAB1 Antibody (HY-P83414) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  Immunohistochemical analysis of paraffin-embedded Mouse testis tissue using JAB1 Antibody (YA3159). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P83414, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
