Description

IFN gamma Antibody (YA4903) is a mouse-derived and non-conjugated monoclonal antibody, targeting to IFN-γ. It can be applicated for WB, ELISA assays, in the background of human.

Background

IFN-γ is a type II interferon produced by immune cells such as T-cells and NK cells, playing crucial roles in antimicrobial, antiviral, and antitumor responses by activating effector immune cells and enhancing antigen presentation. It primarily signals through the JAK-STAT pathway after binding to its receptor IFNGR1, affecting gene regulation. Upon IFNG binding, the intracellular domain of IFNGR1 opens to facilitate association with downstream signaling components JAK2, JAK1, and STAT1, leading to STAT1 activation, nuclear translocation, and transcription of IFNG-regulated genes. Many of these induced genes are transcription factors (e.g., IRF1) that further drive subsequent transcriptional waves. IFN-γ contributes to the class I antigen presentation pathway by replacing catalytic proteasome subunits with immunoproteasome subunits, thereby increasing the quantity, quality, and repertoire of peptides for MHC class I loading. It also enhances peptide generation efficiency by inducing the expression of PA28, which associates with the proteasome and alters its cleavage preference. Additionally, IFN-γ upregulates MHC II complexes on the cell surface by promoting the expression of key molecules like cathepsins B/CTSB, H/CTSH, and L/CTSL. By similarity, IFN-γ also regulates hematopoietic stem cell development, quiescence, and differentiation under developmental and homeostatic conditions.
