C-erbB-2/HER2 Antibody (YA388) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to C-erbB-2/HER2.

Background

ERBB2 is a protein tyrosine kinase that forms part of several cell surface receptor complexes but typically requires a coreceptor for ligand binding. It is an essential component of the neuregulin-receptor complex, though neuregulins do not interact with it alone. GP30 is a potential ligand for this receptor. ERBB2 regulates the outgrowth and stabilization of peripheral microtubules (MTs). Upon activation, the MEMO1-RHOA-DIAPH1 signaling pathway induces the phosphorylation and inhibition of GSK3B at the cell membrane, preventing APC and CLASP2 phosphorylation and enabling their membrane association. Subsequently, membrane-bound APC facilitates the localization of MACF1 to the cell membrane, which is critical for microtubule capture and stabilization. In the nucleus, ERBB2 is involved in transcriptional regulation, binding to the 5'-TCAAATTC-3' sequence in the PTGS2/COX-2 promoter to activate transcription. It also contributes to CDKN1A transcriptional activation via STAT3 and SRC and enhances rRNA gene transcription by RNA polymerase I, promoting protein synthesis and cell growth.

  • Western blot analysis of extracts from A549(lane2(20μg) and A549(lane 3(40μg) using HER2 Antibody (HY-P80500) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Immunocytochemistry analysis of MCF-7 cells labeling HER2 with HER2 Antibody (HY-P80500) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with HER2 Antibody (HY-P80500) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).
