Background

GPER1 is a G-protein coupled estrogen receptor that binds 17-beta-estradiol (E2) with high affinity, triggering rapid transient activation of multiple intracellular signaling pathways. It stimulates cAMP production, calcium mobilization, and Src tyrosine kinase activity, inducing heparin-bound epidermal growth factor (HB-EGF) release and subsequent EGFR transactivation to initiate downstream pathways including PI3K/Akt and ERK/MAPK. This receptor mediates pleiotropic functions across cardiovascular, endocrine, reproductive, immune, and central nervous systems: providing cardioprotection by reducing hypertrophy and perivascular fibrosis via RAMP3; regulating blood pressure through vasodilation and inhibition of vascular cell proliferation; contributing to glucose homeostasis by enhancing pancreatic β-cell insulin secretion; triggering mitochondrial apoptosis during spermatocyte differentiation. Additionally, GPER1 stimulates uterine epithelial proliferation and oxytocin-responsive contractility, induces thymic atrophy via apoptosis, attenuates TNF-driven endothelial adhesion molecules, promotes hippocampal neuritogenesis, and protects against NMDA-induced neuronal death. In neuroendocrine regulation, it increases firing activity and calcium oscillations in LHRH neurons. During skeletal development, it suppresses growth plate osteoblast proliferation and inhibits adipocyte differentiation/lipid accumulation. In cancer contexts, GPER1 drives cancer-associated fibroblast (CAF) proliferation through EGFR/ERK signaling, potentially acts as a transcription factor with EGFR to activate growth genes (c-fos, cyclin D1), and enhances integrin α5β1-fibronectin matrix assembly in breast cancer cells.
