Glypican-3 Antibody (YA5742) is a mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to Glypican-3. It can be applicated for IHC-P, ELISA assays, in the background of human.

Glypican-3 is a cell surface proteoglycan (PubMed:14610063). When attached to the cell surface via GPI-anchor, it negatively regulates the hedgehog signaling pathway by competing with the hedgehog receptor PTC1 for binding to hedgehog proteins (By similarity). Binding to the hedgehog protein SHH triggers endocytosis and subsequent lysosomal degradation of the complex (By similarity). Additionally, Glypican-3 positively regulates the canonical Wnt signaling pathway by binding to the Frizzled receptor and enhancing its interaction with Wnt ligands (PubMed:16227623, PubMed:24496449), as well as the non-canonical Wnt pathway (By similarity). It interacts with CD81, reducing free CD81 available for binding to the transcriptional repressor HHEX, leading to HHEX nuclear translocation and transcriptional repression (By similarity). Glypican-3 also inhibits the dipeptidyl peptidase activity of DPP4 (PubMed:17549790), plays a role in limb patterning and skeletal development by modulating cellular responses to BMP4 (By similarity), regulates the effects of BMP2, BMP7, and FGF7 on renal branching morphogenesis (By similarity), is essential for coronary vascular development (By similarity), and controls cell movements during gastrulation (By similarity).
