Gfi-1 Antibody (YA4878) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to Gfi-1.

Background

Gfi-1 is a transcription repressor essential for hematopoiesis, functioning in a cell-context and development-specific manner. It binds to the 5'-TAAATCAC[AT]GCA-3' sequence in the promoter regions of numerous genes and is a component of several complexes (e.g., EHMT2-GFI1-HDAC1, AJUBA-GFI1-HDAC1, and RCOR-GFI-KDM1A-HDAC complexes) that suppress genes involved in multilineage blood cell development via histone deacetylase (HDAC) recruitment. Gfi-1 regulates neutrophil differentiation, promotes lymphoid cell proliferation, and is required for granulocyte development. By similarity, it inhibits SPI1 transcriptional activity at macrophage-specific genes, repressing macrophage differentiation of myeloid progenitor cells and promoting granulocyte commitment. It also mediates, with U2AF1L4, the alternative splicing of CD45 and controls T-cell receptor signaling (By similarity). Gfi-1 modulates the endotoxin-mediated Toll-like receptor (TLR) inflammatory response by antagonizing RELA and cooperates with CBFA2T2 to regulate ITGB1-dependent neurite growth. In response to TGFB1, it represses CDKNIA/p21 transcription via ZBTB17 recruitment to the CDKNIA/p21 and CDKNIB promoters to control cell-cycle progression. By similarity, it is required for maintaining inner ear hair cells. Additionally, Gfi-1 acts as a substrate adapter for PRMT1 in the DNA damage response, facilitating the methylation of TP53BP1 and MRE11 to promote the DNA damage response.
