DAP kinase 3 Antibody (YA3535) is a mouse-derived and non-conjugated IgG1 antibody, targeting to DAPK3. It can be applicated for WB, IHC-P, FC, ELISA assays, in the background of human.

DAP kinase 3 is a serine/threonine kinase involved in regulating apoptosis, autophagy, transcription, translation, and actin cytoskeleton reorganization. It plays a role in smooth muscle contraction by modulating both type I (caspase-dependent) apoptotic and type II (caspase-independent) autophagic cell death pathways, depending on cellular conditions. It also regulates starvation-induced autophagy and myosin phosphorylation in both smooth muscle and non-muscle cells. In smooth muscle, it directly phosphorylates MYL12B and MYL9 or inhibits smooth muscle myosin phosphatase (SMPP1M) via PPP1R12A phosphorylation, enhancing calcium sensitivity and promoting contraction. In non-muscle cells, it reorganizes the actin cytoskeleton by phosphorylating MYL12B. Isoform 2 can phosphorylate myosin, PPP1R12A, and MYL12B, and its overexpression leads to actin stress fiber bundling. Additionally, DAP kinase 3 is involved in actin filament focal adhesion dynamics, modulated by RhoD. It positively regulates canonical Wnt/beta-catenin signaling through interactions with NLK and TCF7L2. Upon interferon-gamma activation, it phosphorylates RPL13A at Ser-77, triggering its release from ribosomes and association with the GAIT complex for transcript-selective translation inhibition. It also enhances transcription from AR-responsive promoters in a hormone- and kinase-dependent manner and may function as a tumor suppressor by regulating cell cycle progression and proliferation.
