Background

CDC2 is a pivotal kinase that controls the eukaryotic cell cycle by modulating centrosome dynamics and mitotic onset. It drives G2-M transition via interactions with interphase cyclins and phosphorylates substrates such as PARVA/actopaxin, APC, and BARD1 to regulate centrosome separation, Golgi reorganization, nuclear envelope breakdown, and chromosome condensation. CDC2 also orchestrates spindle assembly (e.g., via NEDD1 phosphorylation), microtubule stability, and apoptotic signaling (e.g., activating Bcl-xL/BCL2L1 while suppressing CASP8). DNA damage triggers CDC2 inactivation via phosphorylation, enabling G2 checkpoint arrest; subsequent WIP1-dependent reactivation resumes cell cycle progression. Notably, CDC2 phosphorylates the circadian clock repressor NR1D1 to regulate BMAL1 expression amplitude and inhibits CGAS during mitosis to prevent autoimmune activation. Similar roles include phosphorylating KRT5 (by similar) during prometaphase and CALD1 (by similar) in Schwann cell migration. Additionally, CDC2 serves as a receptor for hepatitis C virus (HCV) entry into hepatocytes.
