CD352 is a self-ligand receptor of the signaling lymphocytic activation molecule (SLAM) family. Triggered by homo- or heterotypic cell-cell interactions, SLAM receptors modulate the activation and differentiation of various immune cells, thereby regulating both innate and adaptive immune responses. Its activity is controlled by the presence or absence of small cytoplasmic adapter proteins SH2D1A/SAP and/or SH2D1B/EAT-2. It triggers cytolytic activity exclusively in natural killer (NK) cells expressing high surface densities of natural cytotoxicity receptors (NCRs). Positive signaling in NK cells involves VAV1 phosphorylation, and activation appears to depend on SH2D1B rather than SH2D1A. In conjunction with SLAMF1, it regulates the transition from positive selection to subsequent expansion and differentiation of thymocytic natural killer T (NKT) cell lineage (by similarity). It promotes T-cell differentiation into a Th17 helper phenotype, enhancing IL-17 secretion, a costimulatory activity requiring SH2D1A. It recruits RORC to the IL-17 promoter. Together with SLAMF1 and CD84/SLAMF5, it may act as a negative regulator of humoral immune responses. In the absence of SH2D1A/SAP, it transmits inhibitory signals to CD4(+) T-cells and NKT cells. It negatively regulates germinal center formation by inhibiting T-cell:B-cell adhesion, likely through increased association with PTPN6/SHP-1 via ITSMs in the absence of SH2D1A/SAP. However, it is also reported to maintain B-cell tolerance in germinal centers and prevent autoimmunity (by similarity).
