Background

Arrestin-β-2 is a multifunctional protein that primarily regulates G-protein coupled receptor (GPCR) signaling by mediating receptor desensitization and resensitization. It binds to GPRK-phosphorylated receptors, sterically hindering their coupling to G-proteins (homologous desensitization). Additionally, as an endocytic adapter (CLASPs), it recruits GPCRs to the AP-2 complex in clathrin-coated pits (CCPs) to promote receptor internalization. The extent of Arrestin-β-2 involvement varies significantly depending on receptor type, agonist, and cell context. It also mediates internalization of non-GPCR receptors such as TGFBR2, LDLR, and ACKR3. Acting as a signaling scaffold, Arrestin-β-2 activates MAPK pathways (e.g., ERK1/2 and JNK3) and AKT1, though the activated ERK1/2 and JNK3 are largely confined to cytoplasmic endocytic vesicles (beta-arrestin signalosomes). It further modulates inflammatory responses, insulin resistance, and inhibitory signaling in natural killer cells. by similar: Arrestin-β-2 and Arrestin-β-1 (ARRB1) exhibit codependent regulation in certain GPCR pathways (e.g., ADRB2, F2RL1, PTH1R), while in others, one arrestin form may dominate while the other inhibits (reciprocal regulation).
