ADH5, a Class-III alcohol dehydrogenase, plays a pivotal role in cellular detoxification processes by catalyzing the oxidation of long-chain primary alcohols and S-(hydroxymethyl) glutathione. Additionally, ADH5 is involved in the oxidation of long-chain omega-hydroxy fatty acids, such as 20-hydroxyeicosatetraenoic acid (20-HETE), yielding both the intermediate aldehyde, 20-oxoarachidonate, and the final product, a dicarboxylic acid, (5Z,8Z,11Z,14Z)-eicosatetraenedioate. Despite its classification as an alcohol dehydrogenase, ADH5 exhibits low efficiency in oxidizing ethanol. Notably, ADH5 is crucial for cellular defense against formaldehyde, a cytotoxic and carcinogenic metabolite, contributing to the clearance of this substance and preventing potential DNA damage.

  Western blot analysis of extracts from MCF-7 (lane 2(20μg), K562 (lane 3(20μg) and HepG2 (lane 4(20μg) using ADH5 (HY-P82253) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
