TP53BP1 is a double-strand break (DSB) repair protein involved in DNA damage response, telomere dynamics, and class-switch recombination (CSR) during antibody genesis. It plays a key role in DSB repair by promoting non-homologous end joining (NHEJ)-mediated repair and counteracting the homologous recombination (HR) repair protein BRCA1. Upon DNA damage, phosphorylation by ATM facilitates its interaction with RIF1 and dissociation from NUDT16L1/TIRR, leading to recruitment to DSB sites. TP53BP1 is recruited to DSBs by recognizing histone H2A monoubiquitinated at Lys-15 (H2AK15Ub) and histone H4 dimethylated at Lys-20 (H4K20me2), which are histone marks present at DSB sites. It is essential for immunoglobulin class-switch recombination (CSR), a process involving DSB generation. Additionally, by similar mechanisms (by similar), TP53BP1 participates in the repair and orientation of broken DNA ends during CSR but is not required for classic NHEJ or V(D)J recombination (by similar). It also promotes NHEJ of dysfunctional telomeres through interaction with PAXIP1.
