Alcian Blue Staining Kit (pH 2.5) is a copper-titanium cyanine conjugated cationic dye that can bind to acidic groups, i.e., form insoluble complexes with anionic groups such as carboxyl groups contained in tissues.

In Vitro

The Alcian Blue Staining Kit, also known as the standard Alcian Blue staining solution, utilizes different pH values ??to distinguish the types of mucus substances. At pH 1, carboxyl groups (COOH) do not stain, while sulfate groups (OSO3H) do. At pH 2.5, carboxyl groups stain well, while sulfated mucus stains poorly, resulting in staining of sulfated mucin and salivary mucin. Neutral mucins (such as those found in the gastric mucosa and Brunner glands) do not react with Alcian Blue.
