Description

Alcian Blue Staining Kit (pH 1.0) is a copper-titanium cyanine conjugated cationic dye that can bind to acidic groups, i.e., form insoluble complexes with anionic groups such as sulfate groups (OSO3H) contained in tissues.

In Vitro

Guidelines (The following are our recommended protocols. This protocol is for reference only and should be modified according to your specific needs.)
1. Dewax the sample with xylene or dewaxing clearing solution, pass it through a gradient of ethanol, and then rehydrate it in distilled water.
2. Prepare the acidification working solution according to the ratio of Alcian acidification solution to distilled water = 1:2, and soak for 5 minutes.
3. Immerse in Alcian staining solution for 25-40 min, then rinse with running water for 5 min.
4. Rinse briefly with Alcian acidification working solution, then blot dry with filter paper to remove excess solution.
5. Counterstain with Nucleotide Red staining solution for 10 min, then rinse with running water.
6. Dehydrate with ethanol as usual, clear with xylene or dewaxing clearing solution, and mount with neutral resin.

Staining Results:
Sulfated mucin appears blue;
Non-sulfated acidic mucin substances do not stain;
Cell nuclei appear light red.

Note:
1. 10% neutral formalin can be used as the fixative.
2. This solution can differentiate between sulfated mucin and proteoglycans; its pH is 1.0.
3. Once opened, the reagent should be used as soon as possible.
