Description

Napsagatran (Ro 46-6240) is a potent thrombin inhibitor. Napsagatran exhibits anticoagulant effects in vitro and in vivo. Napsagatran can be used in antithrombotic research[1][2].

In Vitro

Napsagatran (10 μg/kg/min) reduces platelet-rich thrombus formation in an ex vivo perfusion chamber model[1].

In Vivo

Napsagatran (3-10 μg/kg/min) significantly prevents coronary artery occlusion and reduces intracoronary thrombus formation in a canine model of electrically induced arterial thrombosis[1].
Napsagatran (10 μg/kg/min, i.v., infusion for 240 minutes) significantly reduces thrombus associated fibrin in rabbits[2].

Animal Model: New Zealand rabbits[2]
Dosage: 10 μg/kg/min
Administration: i.v., infusion for 240 minutes
Result: Showed a 1.3-fold prolongation of the prothrombin time (PT) and a 1.7-fold prolongation of the partial thromboplastin time (APTT).
Significantly reduced thrombus associated fibrin after 240 min blood flow exposure.
Showed no effects in heart rate, blood pressure, plasma fibrinogen concentrations, hematocrit and blood cell counts over time.
