Description

VC1052 is a dialkoxypropylammonium-based material and is the core cationic lipid component of the cationic lipid adjuvant Vaxfectin (HY-142998). Vaxfectin consists of VC1052 and lipid DPyPE (HY-112766) in a 1:1 molar ratio. Vaxfectin can be used as a vaccine adjuvant for the preparation of plasmid DNA and protein vaccines[1].

In Vitro

VC1052 has a similar structure to GAP-DLRIE and DMRIE, and is a member of the dialkoxypropylamine cationic lipid class[1].
GAP-DLRIE is primarily used to enhance gene transfection efficiency in local tissues (e.g., lungs), but VC1052 does not enhance pDNA transfection in muscle tissue when it forms a complex with the neutral lipid DPyPE at a 1:1 molar ratio. However, it significantly enhances humoral immune responses (antibody titers increased by 20-50 times) by promoting pDNA uptake by antigen presenting cells (APCs) and inducing cytokine secretion (e.g., IL-6) [1].
GAP-DLRIE significantly enhances the delivery efficiency and gene expression level of pDNA in specific tissues (e.g., mouse lungs) by forming a lipoplex with plasmid DNA (pDNA). GAP-DLRIE relies on cationic lipids to bind to the negatively charged phosphate backbone of DNA to form a stable complex that promotes cellular uptake of DNA and protects DNA from nuclease degradation[1].
